This qualified candidate will be the technical lead for the development of expression and purification methods for recombinant proteins used in the manufacturing of Avitide’s affinity chromatography products. This role includes developing efficient expression systems as well as purification procedures for novel affinity ligands. The position requires working closely with the manufacturing process engineer to ensure an efficient transition for scale-up production. Avitide is positioned to disrupt the cadence of commercialization for novel and high-performance biopharmaceutical affinity chromatography products. This role will enable the development, manufacturing, and supply of these essential technologies to Avitide’s (Repligen) biopharmaceutical partners enabling new and critical medicines that will positively impact human health.
Optimize strain selection, media, growth, culture time, and induction conditions for protein expression in bacterial and yeast expression systems (E. coli or Pichia). Use DOE approach to determine optimal conditions.
Scale-up fermentation to 10-20L scale bioreactor to verify process robustness and transferability.
Develop scalable purification processes that will ensure optimal ligand yield, purity, and activity:
Compare and optimize lysis/extraction procedures (e.g. cavitation, chemical lysis, TFF, centrifugation) to improve recovery while preserving activity
Develop and optimize chromatographic methods (e.g. ion exchange, affinity, hydrophobic, SEC) to obtain required purity while maintaining yield and efficiency
Optimize ligand formulation and storage conditions to ensure integrity
Work in close collaboration with the Manufacturing Process engineer to ensure flawless transition to large scale production
Work in close collaboration with other protein production sites within the company to learn and share best practices
Act as the subject matter expert within the cross-functional product development team. Advise discovery team on expression and purification strategies.
Write and review development reports, transfer documents and other documentation associated with product development and transfer to manufacturing
Manage fermentation and chromatography personnel and assign daily activities.
Provide technical support for current activities to junior personnel
Ensure optimal supply of reagents and chemicals to support uninterrupted activities
Ensure maintenance of fermentation and purification equipment to support activities
Qualifications:Education and Experience:
Ph.D in Bioengineering or Chemical Engineering or equivalent with 5+ years’ experience in a biopharmaceutical environment
Extensive experience with protein expression in E. coli and yeast (e.g. Pichia). Hands-on experience with shake flasks, DASGIP, 10-20 L bioreactors a must
Strong experience with the development of purification procedures for recombinant proteins
Experience with process transfer from R&D to manufacturing a plus
Strong knowledge of MS Office, Minitab (or JMP)
Sig Sigma black belt with DOE experience a plus
Excellent communication skills and ability to thrive in an agile, team-oriented environment
Ability to provide evening, nighttime, and weekend support as needed
